This four-part virtual symposium introduces the theory, design, analysis and real-world application of N-of-1 trials and single-case experimental designs (SCEDs). International experts discuss the history of the methodology, practical study design, statistical analysis, simulation techniques, field development, and applications in clinical practice.

Session 1: Introduction to N-of-1 Trials
Jason Bobe introduces the symposium with the historical origins of N-of-1 trials before exploring how advances in digital health and personalised medicine are transforming the field. He also provides an overview of the symposium and its speakers.
Richard Kravitz explains how N-of-1 trials help address heterogeneity of treatment effects by determining what works best for an individual rather than relying solely on average effects from conventional clinical trials. He discusses the history, design and clinical value of personalised experiments.
Session 2: Design and Analysis Methods
Dr Moeyaert introduces the key principles of single-case experimental designs using a practical sleep study example. She explains common design features, randomisation approaches and methods for analysing repeated-measures data collected within individuals.
Bethany Percha demonstrates how simulation can be used to optimise N-of-1 trial design before a study begins. She explains how simulations help determine sampling requirements, assess statistical power and evaluate different design choices using examples from published studies.
Session 3: Building the Field
Jane Nikles reflects on more than two decades of experience establishing one of the world’s leading academic centres for N-of-1 trials. She discusses the evolution of the field, current research programs and future opportunities for personalised healthcare.
Suzanne McDonald introduces the International Collaborative Network for N-of-1 Trials and Single-Case Designs (ICN), outlining its mission to connect researchers, clinicians and consumers while advancing education, collaboration and methodological development.
Session 4: Patient Community Focus – Persistent Lyme Disease
John Aucott discusses persistent symptoms following Lyme disease, including fatigue, pain, cognitive impairment and sleep disturbance, and explains why personalised approaches such as N-of-1 trials may be valuable in this population.
Richard Schweig reviews the fundamentals of sleep physiology and discusses integrative approaches to managing sleep disturbance associated with persistent Lyme disease.
Emmanuel Varga introduces common sleep disorders, clinical assessment tools and current approaches to diagnosis and treatment.
